Muguruza, the tournament’s ninth seed, defeated Andrea Petkovic 6-4, 6-2. She could face in the next round the 18e seed Victoria Azarenka, who made the final last year at Flushing Meadows.
Jabeur, the 20e seed, easily took the measure of Maria Camila Osorio Serrano 6-0, 6-1 in just 53 minutes.
They were joined hours later by Simona Halep, who reached the third round of the New York tournament for the first time since 2016.
The Romanian crushed Kristina Kucova 6-3, 6-1 at Arthur Ashe Stadium.
Halep, at 12e seeded, won two career Grand Slams, but never excelled at the US Open. She packed up in the first round in 2017 and 2018, and had never passed the second round since her participation in the quarter-finals in 2016.
She triumphed under the retractable roof, as play was interrupted outside due to inclement weather.
Earlier on Wednesday, Japan’s Naomi Osaka advanced to the third round following the withdrawal of her opponent for medical reasons.
The defending champion was due to face Serbian Olga Danilovic in curtain raiser on Wednesday at Arthur Ashe Stadium.
