JAKARTA, KOMPAS.com – Deputy VII of the State Intelligence Agency ( BIN) Wawan Hari Purwanto stated that his party set a higher standard threshold swab test (swab) with the method reverse transcriptase polymerase chain reaction (RT-PCR).

Wawan responded to the news on Tempo Magazine which calls the results of the swab test ( swab test) that BIN did not accurately.

“BIN implements a standard threshold for PCR test results that is higher than other institutions or institutions which is reflected in the value of Ct qPCR. The usual threshold is 35 but to prevent OTG (people without symptoms) from passing screening then BIN raised it to 40, “said Wawan in a written statement, Monday (28/9/2020).

“This includes conducting a validity test through triangulation of three types of genes, namely RNP / IC, N and ORF1ab,” said Wawan.

He said, the difference in swab test results from initially positive to negative is not new.

Because, in the second test, the virus RNA was left very little, even close to missing, so it was not detected.

He said, Covid-19 patients with OTG status could have different results if they were swabbed.

The difference in the results of the swab tests in the near term was also due to the pre-analytic bias.

This happened because the sampling was done by two different people with different quality of training, standard operating procedures (SOP), and different laboratories.

Thus, the swab test sample containing the corona virus was not taken or contaminated.

In addition, Wawan said, reagent sensitivity also determines the results of the swab test.

He said that BIN used Perkin Elmer reagents from the United States (US), A-Star Fortitude from Singapore, and Wuhan Easy Diag from China.

Wawan said the three reagents had a higher level of sensitivity compared to other reagents used in several hospitals.

“Thus, there are several factors that can affect the swab test, including the condition of the equipment, the time of testing, the condition of the patient, and the quality of the test kit,” said Wawan.

He also ensured that the PCR machine used for testing had followed the Bio Safety Level (BSL) 2 laboratory protocol standard.

The machine has also been certified by an international certification body, World Bio haztec.

BIN also collaborates with the Eijkman Institute for Molecular Biology in standardizing test results.

“BIN guarantees the condition of the equipment, methods, and test kits used are gold standard in testing Covid-19 samples. “False negative and false positive cases have been reported in many countries such as the US, China and Sweden,” said Wawan.
