It is the first of its kind to be adopted by one of the major US education authorities.
The Los Angeles Joint School District (LAUSD), the second largest in the United States, voted in favor of the decision at a time when the number of Covid-19 cases in the United States continues to rise, driven by the spread of the contagious delta.
LAUSD-run schools have around 600,000 pupils, of whom around 220,000 can be vaccinated against Covid-19.
The decision taken by LAUSD may set a precedent similar to that of other U.S. education authorities.
In the Los Angeles Joint School District, regular testing of students is already taking place and it is mandatory to wear masks on school grounds, both indoors and outdoors. School staff must be vaccinated.
Under the new rules, all children studying in person must receive the first dose of the vaccine by 21 November. They will need to receive a second dose of complete immunity by 10 January.
Children who reach the age of 12 during the school year will be given 30 days to receive the first dose.
