Gongjindan has been loved for centuries, to the point where it is nicknamed ‘the emperor’s herbal medicine.’ Recently, interest in ‘Yukgongdan’, which adds the prescription of Yukmijihwanghwan (a herbal medicine mixed with Rehmannia glutinosa and Cornus officinalis) to Gongjindan, is increasing. In Oriental medicine, Yukgongdan is evaluated as an herbal medicine that improves the function of the liver and kidneys and is effective for brain health. In fact, studies conducted by Jaseng Oriental Medicine Hospital and UC Irvine School of Medicine Research Institute have proven the effects of Yukgongdan on brain nerve cell regeneration and protection. Meanwhile, Dr. Kim Hyun-seong’s research team at Jaseng Hospital of Oriental Medicine’s Spine and Joint Research Institute (Director Ha In-hyuk) announced that they were the first to prove through cell and animal experiments the effect of Yukgongdan on strengthening immunity by activating immune cells. The paper was published in ‘Heliyon’, an SCI(E) level international academic journal.

The research team used the first-generation anti-cancer drug ‘Cyclophosphamide’, an immunosuppressant, to confirm the immunity-boosting effect of the military complex. First, prior to animal testing, spleen cells were isolated from the spleen of experimental mice, cultured for 6 hours, and then treated with cyclophosphamide and yukgongdan, respectively. In the results analyzed 24 hours later, it was confirmed that the survival rate of immune cells increased in a concentration-dependent manner.

The research team confirmed that the number of T cells, B cells, and white blood cells, which are key elements of the immune system, increased in the spleens of mice administered Yukgongdan after immunosuppression. The size of thymus tissue, which had been reduced due to immunosuppressants, also doubled in size depending on the dose of Yukgongdan. In addition, immune cells derived from spleen tissue, such as CD4+, CD8+, and NK cells, were also more activated. Among them, the number of CD8 + cells increased up to 81.8%.

Yukcomdan also showed the effect of suppressing the death of immune cells caused by immunosuppressants. After administration of Yukgongdan, the expression of BAX protein, which promotes the death of immune cells, was significantly reduced, and BCL-2 protein, which suppresses death, increased. In the case of BCL-2, the expression intensity in both thymus and spleen tissues was activated more than twice.

Cytokines are proteins that regulate inflammation and the balance and response of the immune system, and immunoglobulins are proteins that act as antibodies against viruses and bacteria. As a result of the analysis, both cytokines and immunoglobulins decreased due to immunosuppressants, but then increased again in a concentration-dependent manner due to meat complex. In particular, the expression level of interleukin-10 (IL-10), a cytokine that regulates inflammation levels and immune responses in the body, increased approximately three-fold after administration of Yukgongdan. Dr. Hyun-seong Kim, the first author of the paper, said, “In this study, we were able to analyze the immunity-boosting effect of Yuk-Gong-dan and confirm its potential as an immune-boosting agent.” He added, “The therapeutic effect of Yuk-Gong-dan will be more scientifically proven through detailed research in the future.” “I expect that,” he said.
