Description of the thesis subject

The project is part of the development of strategies to better prevent and control emerging infectious diseases and thus to better prepare for future epidemics or public health crises. Bunyaviruses represent several of the priority pathogens listed by WHO as having the potential to cause future pandemics, including Crimean-Congo haemorrhagic fever virus (CCHFV), Lassa fever virus (LASV) and Rift Valley fever virus (RVFV). These viruses are endemic in southern regions of the world, including today in southern parts of Europe. In this context, it is important to understand the molecular mechanisms of infection.
The project aims to develop approaches to study membrane proteins inside eukaryotic cells using advanced concepts in biochemistry and cell biology. The successful candidate will be involved in the production of membrane proteins by cell-free protein synthesis, and will use cell biology to produce samples, with the final goal of structural studies using NMR.

The research will be conducted at the Institute of Molecular Microbiology and Structural Biochemistry. MMSB is a multidisciplinary Joint Research Unit (UMR) managed, in addition to the CNRS, by the University of Lyon. The objective of MMSB is to understand the molecular mechanisms underlying microbial pathogenesis and to study molecular and macromolecular interactions in structural, dynamic and functional terms. The groups use cell biology and biochemistry approaches in combination with structural biology. The MMSB hosts 11 different research groups, totaling more than 50 members.

The techniques used include biochemical methods including protein expression via cell-free protein synthesis, affinity purification, membrane reconstitution, confocal microscopy and FACS.

MMSB is one of the main contributors to the SFR BioSciences, where it hosts the Protein Sciences Facility, which includes preparative biochemistry on bacterial and mammalian cells, structural studies (X-Ray, EM) and mass spectrometry. It is located in the heart of the Biosciences Campus of Lyon, in the Gerland district.
